Towards precise analysis of the wheat genome: preparation of genomic resources for structural and functional characterization

We have joined efforts from three European countries in order to develop genomic resources for positional cloning, genome organization, structural and functional genomics studies in bread wheat. Until now. two wheat genomic BAG (Bacterial Artificial Chromosome) libraries have been constructed. The first BAG library was constructed using the INRA-French cultivar 'Renan' (1000,320 clones, 7 genome-equivalent and 140 kb of mean insert size, Genoplante Project) and the other BAG library from the reference wheat cultivar 'Chinese Spring' (1200,000 clones and 130 kb mean insert size, INRA/BBSRC Project). As the wheat genome is very complex, we have also conducted attempts to optimise construction of BAG libraries from flow-sorted chromosomes (INRA/Olmoucuniversity project). Successful construction of subgenomic, chromosome-specific and arm-specific BAC libraries represents an important step and will greatly facilitate characterization of the genome of this crop. We have also developed original methods and approaches to study wheat genome organization and to characterize several regions harbouring interesting genes. Genomic resources and tools that we have developed constitute the basis for international collaborations and will help in deciphering the genome of this important crop.
