Lipoxygenase (Lox) activity and lox-1 mRNA accumulation in durum wheat during 3 days early development of light- and dark- grown seedlings were localized primarily in coleoptiles, secondarily in roots and lastly in seeds. Lox activity and lox-1 mRNA accumulation decrease after 4 days growth within the plant. After 9 days growth Lox activities in both coleoptiles and roots decrease markedly. Dormant seeds and all parts of seedlings exhibited 4 isoform bands of Lox activity on polyacrylamide gel with prominent bands in 3 day-old coleoptiles. These results suggest both that the lox-1 gene product is produced specifically during early germination and that induction of lox-1 is correlated with the transient accumulation of a functional Lox enzyme.
