Polymerase chain reaction (PCR)-based DNA markers were developed for the precise and efficient transfer of the cytopiasmic male sterility (CMS) restoring gene Rf-3 into new elite lines. Restriction fragment length polymorphism (RFLP) markers RG140 andRG532 were found to be closely linked to Rf-3. Based on the sequence data (STS) of RG140, pairwise primers were designed to amplify genomic DNA from IR62829A (CMS line) and IR46 (restorer line) to identify polymorphic amplified products between the tworice lines (with EcoRI and Pvull enzyme digestion). Two polymorphic markers, amplified with RG140FL/RG140RL, RG140FL/RB, and RG532 FL/RL were identified. The results demonstrate the utility of STS markers for the transfer of Rf3 through marker-assisted selection.
