Cloning of promoter sequence of aflR gene form Aspergillus oryzae

A 8.3kb DNA fragment containign aflR gene was cloned from genome library of Aspergillus oryzae (A.niger) ATCC 14895. Restriction analysis was carried out by digestion of the DNA fargment with various of restriction enzymes. Then a 420bp SacI/SmaI fragment was subcloned into the pBS II plasmid, according to the restriction map, getting a recombinant plasmid pAP1.1. The sequencing reuslt showed that we had successfully cloned the promoter sequence of aflR gene from A.oryzae. This make it possible to do further researches to reveal the mechanism of the aflatoxin production by Aspergillus.
