In 1983, using the tunnel current phenomenon, the scanning tunneling microscope (STM) was developed. STM featuers high resolution, easy operation, and aerial measurement. However, it has a serious drawback in the cases of biological samples. In order to visualize biological samples, their surface must be coated with conductive material to induce electric current. This coating procedure makes the samples unvital.
