Objective: The acetylcholine expression in hypothalamus arcuate nucleus is detected, which image processing and image analysis is done by the image analysis software. We discuss the combining features of Area Percent of Positive Neuron (APPN) and Relative Intensity of Staining Grey Level (RISGL) in the image quantitative analysis of immunohistochemistry. Methods: With the immunohistochemical method, the expression of ACh in hypothalamic arcuate nucleus in rats is detected. And according to the parameters of APPN, RISGL, etc., we make the image quantitative analysis of IHC. Results: The changes of the total area of positive neurons and APPN show almost the same variation, but the sensitivity of APPN is higher slightly. The results of the average intensity of staining grey level in positive neurons and RISGL are not coincident completely. The former may be biased. The combination of APPN and RISGL reflect the dispersion and aggregation of the products in positive neurons. Conclusions: APPN and RISGL can be more reliable and accurate parameters of the image quantitative analysis of IHC. The combination of APPN and RISGL can not only reflect the expression of positive neurons, but also help analyze its mechanism.
