Optimization of cellulase production of an Endophytic Fungus And The Effect on microbial lipid production

The endophytic fungus marked YY11 can accumulate lipid using cellulose. Effects of cultural conditions on cellulase production of YY11 in liquid state fermentation were investigated for increasing lipid productivity. Various factors were investigated and orthogonal test was used to analyse their effects on cellulase production, the results showed the optimized condition for producing cellulase was: smashed corn stalks 50g, glucose2g, NH4NO3 1.75 g, yeast extract 1.25 g, K2HPO4 1g, MgSO4·7H2O 0.5 g, KCl 0.5g, FeSO4 0.01g, Tween-80 0.4g, CaCl20.2g, 40 mL liquid medium in 250 mL flask, 10% of inoculation and 1000mL distilled water, pH 6.5. Under the optimized condition, the FPA, Cxase and β-Glucosidase were 4.79, 10.04, 12.12 U/mL respectively, about 3.47, 3.98 and 3.90 times of those initial value of 1.38, 2.52, 3.11 U/mL. The lipid productivity reached 1.57g/L, increased by 40.18% compared to the initial of 1.12g/L‥
