Although Poly(ortho esters) matrices have been investigated for controlled release of drugs, microencapsulation with this polymer has not been reported. In this study, we have examined several formulation variables that influence the formation of microcapsules by poly(ortho esters) using 5-fluorouracil (5-FU) as the model drug. The effect of drug concentration, surfacant, sorbitan sequioleate (SSO) concentration, polyvinyl alcohol (PVA) concentration and volume of water phase on actual drug loading, entrapment efficiency, particle size distribution and drug release was investigated.
