We used a hybrid nanoparticle system to investigate the targeting of several CD4 binding ligands conjugate to NCs. We demonstrate that unlike positively charged or neutral LCNPs, a negative surface charge showed dramatically lower nonspecific binding and had preferential bindings to CD4+ T cells when conjugated with CD4 mAbs or its fragments. CD4-targeed LCNPs have great promise for delivery of anti-HlV cure agents, vaccines and gene-modifying oligonucleotide drugs that can be applied in a variety of biomedical areas.
