Conclusions: Skeletal muscle ECM provides a novel model for the aging muscle microenvironment which could help understand causes for decreased muscle healing in elderly individuals. Aged skeletal muscle ECM promotes an increase in macrophage pro-inflammatory phenotype and function as assessed by several metrics. This proves that age-related ECM changes are a contributor to altered macrophage responses. Changes in collagen, hydroxyproline or advanced glycation end product content could be responsible but further mechanistic studies are necessary to confirm this. In vivo validation of altered macrophage responses to aged skeletal muscle ECM are also necessary to confirm in vitro findings.
