We have demonstrated a non-invasive, local delivery vehicle that can elicit functional recovery following ischemic stroke in rats, overcoming the challenges of the BBB in stroke treatment. Controlled release of BDNF was achieved for 21 days in vivo with an encapsulation-free strategy using electrostatic interactions and PLGA nanoparticles. The vehicle itself elicited a short term neuroprotective effect in stroke injured rats, while sustained delivery of BDNF augmented plasticity and subsequently induced long term functional recovery.
