Conclusions: The current study demonstrated that our electrospun polyurethane graft can be tuned to achieve burst pressure comparable to the saphenous vein while increasing graft compliance. It is expected that this increased compliance matching will improve long term patency by reducing the onset and severity of intimal hyperplasia. We have also identified an improved hydrogel composition for the inner layer with improved suturability that maintains the desired thromboresistance and cell behavior. Overall, these studies represent an important milestones in the development of a multilayered vascular graft. Future studies will utilize these new grafts in bioreactor models to elucidate the effect of compliance-mismatch on smooth muscle cell phenotype changes related to intimal hyperplasia.
