This paper integrated size-based filtration and negative depletion (SF-ND) to improve the rare circulating tumor cells enrichment from the undiluted whole blood. The whole blood was firstly filtrated by a carefully designed micropore array filter. The captured cells were then released off and mixed with CD45 Dynabeads. CD45+ white blood cells were removed by a magnet and left target cells with a high purity. All operations were conducted in a home-made gadget. The preliminary performance of the SF-ND system showed a high purity (80.48 ± 0.87%) along with the acceptable recovery rate 57.83 ± 10.62% by treating 1 mL whole blood with spiked 104 lung cancer cells (A549s).
