This study evaluated whether improving remodeling of MI hearts and the potentials of angiogenesis by secreting paracrine factors by implanting silk fibroin (SF)/chitosan-hyaluronic acid (HA) (SCH) or SF/ HA (SH) patches onto myocardial infarction (MI) rat hearts induced by a cryo-injury technique. The evaluations of SCH and SH patches on attenuating the remodeling of MI hearts were determined after two months of MI induced. The results showed that the implantations of SH and SCH patches could significantly (P0.05 or better) reduce dilation of LV inner diameters, increase wall thickness of MI hearts and improved LVFS (fractional shortening) values compared with those of cryo-injury group. For instance, the wall thickness of MI hearts of SH, SCH and cryo-injury groups were 1.70.10 (n=8), 1.50.13 (n=8) and 1.20.08 mm (n=8), respectively. Moreover, increasing capillary-like structures or angiogenesis potentials in MI zones for SCH and SH groups were found while those did not happen in the cryo-injury group. Notably, SH group significantly increased wall thickness of MI rat hearts and promoted secreting paracrine factors such as VEGF, bFGF and HGF in MI hearts (P<0.05 or better, n=4 for each group, respectively), determined by real-time PCR, compared with those for SCH group, indicating that SH patches would be better than SCH ones for improving LV remodeling and stimulate various paracrine factors to promote the recovery heart functions of MI rat hearts.
