In summary, the insulin release in vitro could be tuned to obtain desired "on-off" sensitivity in response to stepwise glucose challenge, and the in vivo experiments indicate that the sustained release time of insulin, as well as the hypoglycemic effect, could be prolonged from 17 to 36 days, just by rearranging the assembly sequence of LbL building blocks.
