The use of solid-in-oil nanodispersions for transcutaneous approach has been reported by others. Here, we extend to develop this concept to increase the efficiency of transdermal delivery. The combination of gold nanorods with a solid-in-oil (SO) dispersion to enhance the permeation of the large macromolecule protein (FITC-ovalbumin; MW: 45 kDa) through the mouse skin in vitro was investigated in our study. The exhibition of a strong fluorescent image in mouse skin sections under fluorescent microscopy indicated that the penetration of a model protein (FITC-ovalbumin) into mouse skin significantly increased after treatment with gold nanorods in SO dispersions followed by near infrared light irradiation for 20 min and farther continued incubation for 12 hr. This investigation would provide a new paradigm to improve transcutaneous delivery of macromolecule proteins.
