Penicillium decumbens JU-A10-T is a cellulase-producing fungus stored in our laboratory and can produce cellulase with a suitable enzyme system. Ion beam implantation technique is a new breeding method, and has been used in the breeding of a variety of crops, insects and microorganisms such as Aspergillus oryzae and Aspergillus niger. To improve cellulase productivity and obtain cellulase with high activity of FPase and β-glucosidase, a mutant 140-12 was derived from P. decumbens JU-A10-T by N+ Implantation. This mutant was cultivated by liquor fermentation in a shaker at 180 rpm, 30oC for 120 h, and crude cellulase with filter paper activity of 7.7 U/ml, which was 22% higher than that from strain JU-A10-T, was obtained by filtration and centrifugation. The crude cellulase was used to hydrolyze corncob residue to evaluate its application in enzymatic degradation of cellulosic substrate.
