CHITOSAN/PVA FILM FOR OCULAR DRUG DELIVERY OF WATER SOLUBLE ANTIMICROBIAL AGENTS

The purpose of this study is to development of ofloxacin film for ocular drug delivery, using bioadhesive cationic polymer, chitosan and polyvinyl alcohol (PVA). The film can readily swell in presence of lachrymal fluid; increase the overall residence time of formulation, sustained release of drug possible. Solution of chitosan in lactic acid and PVA in water was prepared by overnight stirring. Two solutions are mixed on dry weight bases in different ratios to give chitosan: PVA blends (1:0, 1:5, 1:10, 1: 15, 1:20, 1:30 and 0:1). The drug was mixed uniformly in prepared blends allow casting in petridish and dried for 48 hours at room temperature to give homogenous films. The films were evaluated for mechanical properties, swelling behaviour and invitro drug release studies. The result of mechanical properties shows that PVA has a positive effect and drug loading has a negative effect on both tensile strength and percentage elongation. Similarly swelling behaviour at neural pH suggests that PVA, which more hydrophilic polymer has a positive effect; also supported by highest drug release observed from plain PVA film (60 % in 1st hour). All film shows more than 90% of drug release within a period of 4 hours. The prepare Chitosan/PVA film in ratio of 1:10 and 1:15, with good mechanical and release characteristic (controlled) can be successfully utilize for ocular drug delivery of antimicrobial agents.
