Based on our novel finding of spontaneous pore closing in PLGA, we have disclosed a new paradigm to microencapsulate drugs, particularly proteins, without subjecting them to destabilizing manufacturing stresses. This process was utilized to create microspheres with 1-6% (w/w) protein loading with minimal initial burst. One-month controlled release was also demonstrated.
