The use of detailed models for bioprocess design and control has been limitedas accurate estimation of model parameters is often difficult. In this paper, theparameter estimation problem for a mechanistic model of the production of abiopolymer, poly-β-hydroxybutyrate (PHB), is examined in detail. Parameterestimation was undertaken using previously published data. Three parametersets were obtained with large uncertainty in some parameters. Parametricsensitivity, identifiability and estimability analyses indicated that only certainparameters were uniquely identifiable. Experiment design studies indicated thatcarefully designed experiments could significantly reduce the uncertainty in theparameter estimation problem.
