For selective and rapid isolation of high-affinity antibodies to any target protein, large phage-displayed naive antibody libraries derived from human, mouse and rabbit, were prepared by using our originally modified PCR primers for amplifying naive single-chain Fv gene. The size of our obtained phage-displayed naive antibody libraries had more than 10~9, respectively. As a result, without in vivo immunization of antigens, many high-affinity antibodies were selectively obtained to various antigens and their epitopes within 1 week by using these libraries. We are currently undergoing to isolate useful high-affinity antibodies for various targeting therapies.
