Altheough cellobiose dehydrogenase (CDH) is capable of reducing a wide range of substrates, including transition metaal ions, quainones, and phenoxy radicals, its true in vivo role has remained maysterious. In order to study the potential imporance of CDH to lignocellulose degradation and pulp biobleaching, we constructed a CDH-deficient strain of the polypore basidiomycete Trametes veriscolor. This strain was developed by transforming protoplasts of T. versicolor strain 52J with a plasmid containing a distrupted version of cloned T. versicolor cdh and a phleomycin resistance casette.
