Effect of Cross - Linking of Agarose on Enzyme Immobilization

摘要

Penicillin amidase is immobilized on cross - linked agarose and noncross - linked beaded agrose gel by means of NalO_4 activation and 1,4 - butanediol diglycidyl ether (bisoxirane) activation. The activities of the four resulting immobilized enzymes (IMEs) are all high enough for industry use. In the case of NalO_4 activation, the IME based on noncross - linked agarose have a much higher activity than that of the IME based on cross - linked agarose. While in the case bisoxirane activation, the IME based on either kind of agarose is has an activity quite close to each other and the recovery of enzyme activity is about 100 %. Discussions are made on reactors and practical use.
