AMP-Activated Kinase (AMPK) is a nutrient and energy sensor that regulates energy homeostasis. Metformin, an activator of AMPK, improves fertility and metabolic profile in women with Polycystic ovarian syndrome (PCOS). PCOS, a common cause of infertility in women, is associated with metabolic disorders. We hypothesize that the positive effect of metformin on reproduction is through its action on AMPK in GnRH neurons to regulate KISS1R, and GNRH1 expression. Our objective was to measure Ampka, Kisslr, and Gnrhl mRNA expression in GT1-7 cells, a murine GnRH neuronal cell line, following treatment with metforming or an AMPK agonist (5-aminoimidazole-4-carboxamide ribonucleoside -AICAR).
