To investigate the effects of autocrine motility factor (AMF) on phosphorylation level of confilin in esophageal squamous cell cancer (ESCC) cell line EC9706. Two recombinant vectors containing full length of AMF cDNA were transfected to EC9706 by Lipofectamin?2000. The location of EGFP-AMF protein was observed under fluorescent microscope. The expression level of AMF in EC9706 cells was examined by RT-PCR and Western blots. The effects of AMF on the expression of phosphorylated cofilin in EC9706 cells were also investigated by Western blots. The Results showed that EGFP-AMF fusion protein was detected mainly in the cytoplasm of EC9706 cells. The protein of AMF was detected only in EC9706 cells transfected with the pcDNA3. l( + )-AMF. The expression levels of phosphorylated cofilin proteins in the EC9706 cells transfected with and without pcDNA3. 1 ( + ) both were much lower than that in EC9706 cells transfected with pcDNA3.1(+)-AMF. It is concluded that AMF is located in the cytoplasm and increases the phosphorylation level of cofilin in EC9706 cells.
