MICROSATELLITE MARKERS FOR CULTIVAR IDENTIFICATION IN SUGARCANE (SACCHARUM SPP.)

Microsatellites (or simple sequence repeats, SSRs) are a powerful tool for classifying sugarcane cultivars and related species. We have used SSR markers to classify a total of 32 sugarcane clones (S. spp. hybrids: 7, S. spontaneum: 17, S. officinarum: 1,S. barberi: 1 and S. spontaneum x S. spp. hybrids: 6), including the Japanese wild S. spontaneum collections. A total of 27 SSR oligonucleotide primer pairs was tested and several primer pairs were selected for further analysis of sugarcane genotypes. Twenty primer pairs were designed according to Saccharum genomic DNA sequence data in GenBank, and another seven primer pairs were the same as those used by Cordeiro and others. Polymorphic markers allowed the identification of most of the sugarcane clones tested, even though some of them are closely related and difficult to classify by morphological characters. Our results also indicate SSR detection by polyacrylamide gel electrophoresis and silver staining is superior to agarose gel electrophoresis and ethidium bromide staining, or radio-labelling and autoradiography. Microsatellite markers can be used for routine, rapid identification of sugarcane clones.
