The controlled release of DNA from single PLGA microspheres has been characterized by measuring the size, weight and DNA content of individual microspheres. DNA distribution within a single PLGA microsphere was evaluated by confocal microscopy; real-time DNA release from individual microspheres was observed and followed using microscopy and digital image analysis. DNA release profiles from individual microspheres with different sizes will be presented. These results will provide insights into the behavior of individual microspheres, and therefore the overall performance of microspheres as DNA delivery systems.
