Autologous Extracellular Matrix Scaffolds Prepared by a Template Method

Biomimetic scaffolds for tissue engineering and regenerative medicine have been developed from natural polymers such as collagen, fibrin, hyaluronic acid and acellular matrices derived from decellularized tissues and organs. Although the naturally-derived polymer and acellular matrices can provide abundant informational signals and degrade into physiologically tolerable degradation compounds, they are exhaustively allogeneic or xenogeneic, which takes the risks of pathogen transmission and may provoke undesirable inflammatory and immunological reactions and lead to the ultimate failure of regenerated tissues and organs. Development of autologous ECM scaffolds (aECM) has been strongly expected for tissue engineering and regenerative medicine. In this study, we developed a novel method to prepare aECM scaffolds by 3D culture of cells in a template that can be selectively removed after cell culture.
