Background: There are a large number of hematopoietic stem cells in murine fetal liver of pregnancy of 14.5day, which have high capacity of self-renewal and proliferation compared with one in bone marrow. The propose will be put forward to that the plasticity of them will be larger than stem cells in bone marrow.rnOBJECT: To explore the capacity of differentiation of Sea-1+ cells from murine fetal liver into renal tissue and cells.rnMETHOD: The single cell suspension was prepared of murine fetal liver of pregnancy of 14.5day; cell sex was determined by quick PCR and male cells were collected, and Sca-1+ cells of them were separated with MACS technology. 2×104 cells of male Sca-1+ cells were transplanted into female syngeneic mouse irradiated with lethally dose of γ ray by tail vein. 2 months late, the kidneys were taken out and paraffin slices of them were made. Furthermore, In order to reduce the interference of female hematopoietic stem cells in vivo, female bone marrow would be changed into male one with elimination of female bone marrow by radiation and transplantation of male Sca-1+ cells from fetal liver, which needed time of 8 weeks. The acute renal failure model of female mice with this male bone marrow and normal female mice were made again, and 72 hours later, Sca-1+ cells from fetal liver were transplanted into them. 8 weeks later, the mice were killed and kidneys were took out to be made tissue slices for detecting of FISH and immunochemistry,rnRESULTS: The male cells were found on sections of kidneys from mice irradiated with lethally dose of y ray, frequency of them was (4.5+0.5)% there appeared a double male cell arrangement on area of renal tubules; (8.58+1.34)% on sections of kidneys from mice with acute renal failure and (18.13+1.91)% on sections of kidneys from mice with acute renal failure and bone marrow change from female into mal mice and there appeared ring arrangement of several male cells on area of renal tubule. It was found with immunochemistry method that the phenotype characteristics of male renal tissue was RCA+/CYP1a2-/Vimentin+/CD45-/F4/80-, which show characteristics of renal tissue.rnConclusion: The Sca-1+ cells from fetal liver have the capacity differentiated into kidneys; acute renal failure can promote the differentiation process.
