Separation of 4-[(4-dimethylamino)-1-(4'-fluorophenyl)-1-hydroxy-1-butyl]-3-(hydroxymethyl)-benzonitrile(diol),from 3-[(acetyloxy)methyl]-4-[4-(dimethylamino)-1-(4-fluorophenyl)-1-hydroxybutyl]-benzonitrile(diol acetate)by solvent extraction was investigated. The partition of diol and diol acetate in variousextraction systems were investigated. Succine anhydride was selected as the derivative agent for diolderivatization. Water/benzene two-phase solvent system was screened as the optimal extraction system for theseparation of diol derivative and diol acetate. Furthermore,the effects of pH of aqueous phase and initialconcentration of diol acetate were studied to improve the efficiency of extraction. The partition coefficient ofdiol acetate reached 157.7 under optimal extraction condition as pH 8,10 mmol/L and 25℃. After threeextraction stages,the purity of diol derivative in the aqueous was 99.2%(mol)and the yield reached 94.3%.
