The present invention relates to a FRET biosensor that targets the binding of a coronavirus family-derived spike protein to ACE2, and a method for screening a spike protein and ACE2 binding inhibitor using the biosensor. The RBD of the biosensor according to the present invention specifically binds to ACE2 in the cell, and whether or not binding and the degree of binding can be measured through a change in the ratio of fluorescence intensity based on the FRET phenomenon, which inhibits RBD-ACE2 binding. It may be usefully used as a means for screening substances, furthermore, substances for preventing or treating coronavirus infection and verifying their efficacy.
