The present invention relates to an enzyme-catalyzed step for producing GDP-fucose from the low cost substrates guanosine and L-fucose in a single reaction mixture, or from guanosine and D-mannose. The steps can be operated (semi) continuously or in batch fashion. In addition, the steps can be adapted to produce fucosylated and biomolecules containing glycans such as, for example, breast milk oligosaccharides, proteins, peptides, glycoproteins or glycopeptides.
