FIELD: biotechnology.;SUBSTANCE: invention relates to the field of biotechnology. Tubercles of gladiolus hybrid are subjected to two-stage sterilization. The tubers are placed in a solution of neutral detergent and incubated for 4 minutes, stirring occasionally. Then the tubules are washed with cold running water for 5 minutes. In a laminar flow hood, 0.02-0.06% sodium hypochlorite solution is poured and kept for 8 minutes with periodic stirring and then placed in 96% ethanol for 10 seconds. Rinse 3 times for 3 minutes with sterile bidistilled water. As an explant, the extracted apical renewal bud is used, which is placed on a sterile agar modified Murashige-Skoog culture medium (MS) with the addition of NAA 2 mg/l and 6-BAP 2 mg/l in Petri dishes. Petri dishes are kept in a room with specified light (16 h - day; 8 h - night) and temperature (21-22°C) conditions. Every 30 days, explants are plated on fresh MS medium as described above. Transferred 3-4 times to a nutrient medium without adding growth regulators 6-BAP and NAA.
