A method of calibrating a darkfield imaging microscope setup is disclosed. The method comprises preparing a plurality of particle samples, each sample having a known concentration and particle size, the plurality of particles having one or more particle sizes, optionally having one or more refractive indexes and one or more diluents. For each sample of the plurality of samples, the samples are measured in that setup and the scattered light intensity and number of particles are measured. From this data, scattered light intensity, particle size and calibrated irradiated volume can be determined. The calibrated irradiated volume can be used to obtain an appropriate particle size distribution for a given diluent.
