The present invention relates to a novel polypeptide designed using a computational biological method to bind to a specific site of domain 4 of human epidermal growth factor receptor 2, a method for preparing the polypeptide, and a use of the polypeptide. Serving as a new diagnostic and therapeutic agent for cancer, the novel polypeptide according to the present invention is expected to substitute for conventional antibodies.
