The present invention relates to a discovery of an activity of esterase of a recombinant spike protein of the Coronavirus Disease 2019 (COVID-19) virus and to development of a treatment for the COVID-19 virus using same. Specifically, the present inventors confirmed for the first time that the S1 subunit and the S2 subunit of the spike protein of the COVID-19 virus each showed high esterase activity, and have conceived a method for developing a therapeutic agent for COVID-19 in such a way that a substance inhibiting the activity of the esterase is selected, on the basis of the activity of the esterase of the spike protein of the COVID-19 virus. In addition, the present invention provides a substance inhibiting esterase, selected through the above method, as a therapeutic agent for COVID-19. The selected substance inhibits cell infection by the virus and a release process of the virus from the cell by inhibiting the activity of the esterase of the spike protein of the COVID-19 virus, thereby inhibiting the proliferation of the virus in a body, and thus has an effect of treating COVID-19.
