The present invention relates to a recombinant protein-producing cell line and a method for preparing the cell line by applying site-specific integration (SSI) and recombinase-mediated cassette exchange (RMCE). By targeting the expression hot-spot, which is known to have high expression level, the RMCE landing pad is inserted through SSI and expressed through the promoter of the hot-spot, resulting in stable and high level of expression. Enables protein production. In addition, since the GOI of a protein-producing cell line can be easily replaced through RMCE, the type of protein produced can be changed while maintaining the protein production of the existing cell line. This can greatly reduce the time and cost involved in the cell line development process in the recombinant protein production industry.
