An optical path position calibration method, a calibration tool used in the method, and a fluorescence quantitative detection system. The optical path position calibration method comprises: dividing optical fibers at a detection end into M optical fiber bundles according to an arrangement mode of M columns and N rows of sample holes, each optical fiber bundle comprising N optical fibers (S101); installing the M optical fiber bundles to enable the position of each optical fiber bundle to correspond to one column of sample holes (S102); and lightening the optical fibers corresponding to the N rows or N-1 rows of sample holes according to a set sequence by using an optical path position calibration tool, and detecting the positions of the N optical fibers of each optical fiber bundle by using a detection device (S103). During multi-hole detection and high-throughput detection using a large number of optical fibers, the positions of a large number of sample holes can be easily and quickly aligned in one-to-one correspondence to the positions of a large number of optical fibers at the detection end, so that the detection result is accurate, and disorder is avoided.
