The present invention provides a method for isolating a sequencing complex.The method forms a complex between a nanopore covalently linked to the polymerase and an oligonucleotide bound to the purified portion; A solid support that can be bonded to the purified portion is used to separate any unbound / non composite nanopores and oligonucleotides from the complex and disconnecting the bond complex from the solid support using the enzyme composition.Diagram
