The present invention includes methods and compositions for converting regulatory T cells (Tregs) into Ten-Eleven Translocation (TET) deficient Treg cells (ex-Treg) in vitro comprising: isolating Tregs from a subject; and culturing the Tregs with an agent that inhibits the activity or expression of one or more TET enzymes in the Tregs, wherein the loss of TET enzyme function converts the Tregs into ex-Tregs.
