The present invention relates to the technical field of genetic engineering, and provides a recombinant E. coli system, a construction method therefor, and an application thereof in the synthesis of an α-1,2-fucosylated oligosaccharide. The recombinant E. coli system has an α-1,2-fucosyltransferase gene and a GDP-fucose pyrophosphorylase gene. By transforming the α-1,2-fucosyltransferase gene and the GDP-fucose pyrophosphorylase gene into E. coli, a genetically engineered strain is constructed to express an α-1,2-fucosyltransferase and a GDP-fucose pyrophosphorylase, and the expressed α-1,2-fucose pyrophosphorylase and GDP-fucose pyrophosphorylase are used for enzymatic synthesis of an α-1,2-fucosylated oligosaccharide.
