The present disclosure relates generally to differentiated tendon cells derived from pluripotent stem cells and methods of making and using such cells. In one aspect, provided is a method of generating a population of tenocytes comprising: (a) culturing multipotent mesoderm in serum-free medium in the presence of transforming growth factor P (TGFP) and optionally one or more of fibroblast growth factor (FGF), sonic hedgehog (SHH), smoothened agonist (SAG), Noggin, Activin A, growth differentiation factor 7 (GDF7), CD1530, and LDN-193189 to produce tenocytes, and (b) optionally isolating the tenocytes.
