2-SUBSTITUTED 1,4-BENZODIOXANES

I compounds of general formula: (I) R, R1 and R2 = halogen, (1-6C) alkyl, or (1-6C) O-alkyl n = 2-6 (integral). II Acid addn. salts and pharm. prepns. of I. Calming, hypnotic and hypotensive drugs for oral, intravenous, or subcutaneous administration. Dose 0.1 to 10 mg./kg. (I) will prevent the blood pressure increase normally caused by stimulation of the splanchnic nerve, or injection of adrenalin or nor-adrenalin. 2.5to 10 mg./kg. subcutaneous in conscious rabbits prevented an increase in blood pressure when noradrenalin was injected. I (R1 = MeO) are particularly effective. 2-Aminomethyl-1:4-benzodioxan (17 g.) and phenoxyethyl chloride (7.83 g.) were refluxed 4 hrs. at 150 deg.C, cooled, CHCl3 (30 ml.) and K3CO3 (7 g.) in H2O (20 ml.), added. Separated, H2O extd. (2 x 10 ml. CHCl3), CHCl3 dried and distilled, giving 2-phenoxyethylaminomethyl-1:4-benzodioxan (70 g.) as a light-brown viscous oil b.p. 204-8 deg.C/1 mm.
