Iodine-labelled L-asparaginase, particularly L-asparaginase-131I, with iodination degree of 0.1-10 (0.5-2) I atoms/molecule, suitable for tumour-diagnosis, is prepared by reacting a buffered L- asparaginase solution in water at pH 7-10 either with (a) I2 (131I or 125I) or I(131I) Cl or I(125I)Cl, or (b) an isotope- containing iodide in presence of an oxidation agent or (c) an isotope-containing iodide under electrolytic oxidation conditions, and separating the labelled L-asparaginase by dialysis or chromatography.
