Plant tissue prodn - by culture in nutrient medium with reduced sugar content

A modified strain of plant-tissue is prepd. by culturing undifferentiated plant-tissue in a 1st nutrient medium contg. a sugar, in the presence of light and CO2. A part of the growing tissue is removed and further cultured in a 2nd nutrient medium contg. less sugar. A non-toxic, non-utilizable substance is added, pref. mannitol, sorbitol or other sugar alcohol, to the 2nd medium in amt. sufficient to equilibrate the osmotic pressure of both media. The light intensity during growth is kept at 2600-10000 Lux, pref. 5500-10000 Lux. The CO2-concn. of the air is increased during growth. Further portions of the growth in the 2nd medium maybe transferred to a 3rd medium which again contains less sugar than the 2nd medium. This may be repeated many times until only traces, or no sugar is present. Used for culturing tissue of grasses, lucern, clover, vegetables and plants such as tobacco.
