Method of obtaining the quantitation and concn. of precipitin reaction participating molecules (I) (e.g. antibodies or antigens) in a biological specimen comprises forming a gradient of reference reactant carrying gel formation, calibrating the gradient by dividing it into zones having different concns. of reactant, placing the biological specimen in contact with the reactant in each of the zones and relating the visible precipitin reaction that occurs to the concn. of reactant in each zone to determine the concn. of (I) in the biological specimen.
