Isolation of biologically active protein - with influence on primary carbohy-drate metabolism and the urea cycle and used to treat uraemia inflammation and shock
A biologically active protein is isolated, esp. from liver tissue, by (a) treating the starting material with aq. soln. of divalent activating metal carbon at neutral or slightly alkaline pH and sepg. the solids; (b) treating the liq. with a heavy metal precipitant and sepg. the ppte.; (c) adjusting the pH of the liq. to 8.3-8.5 and sepg. the ppte.; (d) treating the liq. with a conc. soln. of a salting-out agent and sepg. the ppte.f (e) diluting the liq. with a solvent to give a solvent concn. of 20-26 vol.%, sepg. the ppte., diluting to a solvent concn. of 35-40 vol.% to give a 3-layer mixt., and diluting the upper layer to a solvent concn. of 70-77 vol.%; (f) sepg. the protein ppte. and dispersing it in a solvent with a pH of 7.8-8.2; (g) dialysing the dispersion; (h) heating the dialysed prod. to 46-50 degrees C. and sepg. the ppte.; and (i) subjecting the liq. to electrophoresis to isolate the biologically active protein.
