Colour reagent for quantitative sugar chromatography - by borate complex ion exchange, contains di:sodium bi:cinchoninate, sodium carbonate, aspartic or citric acid and copper sulphate
Dye reagent for the automatic quantitative borate complex ion exchange sugar chromatography contains, by wt., in aq. soln. having pH 11.2-11.7, 0.04-0.25 % di-Na 2,2'-bicinchoninate, (I), 2-7% Na2CO3, 0.05-0.2% aspartic acid or citric acid monohydrate and 0.02-0.15% CuSO4 (calculated as the pentahydrate). The reagent is pref. used as 0.5-5 ml aq. soln./ml. soln. leaving the chromatography column. The reagent can be used for most reducing sugars, e.g. hexoses, pentoses, amino-sugars, Me ethers, di- and oligosaccharides, glucoronic acids, etc. The reagent is non-corrosive and highly sensitive, e.g. down to 0.01 mu g. The colour reagent has an intense blue colour and strong fluorescence. When stored in brown bottles at room temp., discolouration sets in only after several weeks and the reagent acquires a purple shade. The formation of coloured complexes with reducing sugars is constant however and unchanged for several months.
