(III) is prepared by condensing ethyl (2-methyl-4,5-dimethoxybenzyl)cyanoacetate or diethyl(2-methyl-4,5-dimethoxybenzyl)melonate with guanidine or urea to give (I). The free amino groups are protected by acetylation and the protected (I) chlorinated with POCl3 or PCl5 to give (II) which is then aminated and catalytically dehalogenated with H2 in the presence of Pd/C to give (III) (where Z'=2-methyl-4,5-dimethoxybenzyl; R'=CN or CO2-Et; X and Y = same or different OH or NH2; Z and W = same or different Cl or NH2).
