STABILIZING METHOD OF REAGENT FOR DETECTING RHEUMATOID FACTOR

PURPOSE:To suppress the time aging change of reagent and to make stable without self agglutination due to freezing and thawing, by adding specific amount of choline bromide or iodide to the latex particle suspension sensitized with human gamma-globulin. CONSTITUTION:To the 5% (W/V) polystyrene latex solution, one volumen, 1% (W/V) human gamma-globulin solution, 4 volumes are added and after heating at 37 deg.C temperature for one hour, it is cooled and left at a cold place for a night. Next, the centrifugal separation is made, the precipitate is floated in the 0.1M glycin buffer solution to obtain the latex concentration of 0.5-1.0 (W/V), and after heating for increasing sensitivity, the reagent is prepared by adding choline bromide or iodide by 1-40wt%. After the human gamma-globulin sensitized latex reagent 0.05ml obtained is mixed on the slide glass with the inspected body secrum, 0.05ml, the degree of agglutination is observed standstill. Thus, the stable reagent can be obtained, which has no change in the reaction even at the storage for one yeat at 2-10 deg.C and at the thawing after freezing at -20 deg.C temperature.
